Properties of Capsular Polysaccharide Deolymerase Associated with Phage-infected Klebsiella pneumoniae

Abstract
The capsular polysaccharide depolymerase produced by phage-infected Klebsiella pneumoniae type 2 was isolated as a soluble form, and some of its properties were studied using an assay method based on determination of a reaction product.
Exposure of capsular polysaccharide containing 40% neutral sugars to the enzyme resulted in a measurable increase in the level of acetone-soluble neutral sugar. The liberation of the neutral sugar from the substrate by the enzyme was most active at pH 7.5, and proportional to some extent to the incubation time and to the enzyme concentration. The crude enzyme showed a remarkable heat-stability, being not inactivated by heat-treatment at 60¡Æ for 20 minutes.
These results indicate that the enzyme is similar to the other depolymerase of phage-infected Klebsiella-Aerobacter strains in releasing detectable amounts of neutral sugars from capsular polysaccharide.
